ExperimentMic multiplexed FISH on F123-CASTx129 with Sox2 tags and RMCE site downstream of Sox2 SE

Imaging the 7.5kb insert in a different location (downstream of the super enhancer) to test whether insulator function is dependent upon the precise postitioning of the insert relative to the Sox2 gene and super enhancer. We also colocalized the chromatin data with nascent RNA images to understand bursting behavior as a function of the allele classification.
